Role of CST1 in Promoting Gastric Cancer Metastasis and Analysis of Its Mechanism

Objective: To elucidate the role and mechanism of cysteine protease inhibitor SN (CST1) in promoting metastasis gastric cancer. Methods: (1) Firstly, 6 pairs cancer paracancer tissue samples from patients without distant 5 with peritoneal were collected for transcriptome sequencing. Statistical analysis was performed on sequencing results to identify significantly upregulated differential genes. (2) Another 75 patients, protein total RNA extracted. Western blot quantitative reverse transcription polymerase chain reaction (qRT-PCR) used detect expression CST1 messenger (mRNA) adjacent tissues. (3) The AGS, BGC823, HGC-27, MGC803, MKN45, SGC7901, SNU-1 cells normal mucosal epithelial GES-1 qRT-PCR mRNA expression. Results: gene screened, tissues higher than that Compared GES-1, cell lines upregulated, HGC-27 relatively low, while AGS MKN45 high. At same time, a stable line overexpressing knocking down successfully constructed. Overexpression or knockdown did not change proliferation ability cells, but can promote migration invasion cells. (4) may by activating epithelial–mesenchymal transition (EMT) signaling pathway. Conclusion: promotes we found through animal experiments affect function is negatively correlated level E-cadherin.
